Overview
Sirexatamab + atezolizumab is a combination immunotherapy regimen being evaluated for the treatment of advanced gastroesophageal cancers. Sirexatamab (DKN-01) is a humanized IgG4 monoclonal antibody that binds to and neutralizes Dickkopf-1 (DKK1), a protein that inhibits the canonical Wnt signaling pathway and is associated with an immunosuppressive tumor microenvironment and poor prognosis. Atezolizumab is a humanized IgG1 monoclonal antibody that targets programmed death-ligand 1 (PD-L1), preventing its interaction with PD-1 and B7.1 receptors to restore anti-tumor T-cell activity. The combination is specifically being studied in the Phase II WaKING trial, sponsored by the Royal Marsden NHS Foundation Trust, targeting patients with mismatch repair proficient (pMMR) or microsatellite stable (MSS) oesophagogastric adenocarcinoma who have progressed on prior chemotherapy.
