Overview
Sirexatamab + bevacizumab + chemotherapy is an investigational combination regimen being developed for the treatment of advanced colorectal cancer. The regimen's primary novel component is sirexatamab (DKN-01), a humanized monoclonal antibody that targets Dickkopf-1 (DKK1), a secreted protein that modulates the Wnt signaling pathway and is associated with tumor progression and an immunosuppressive microenvironment. Bevacizumab is a standard-of-care anti-angiogenic antibody targeting vascular endothelial growth factor (VEGF). The chemotherapy component typically consists of standard regimens such as FOLFOX or FOLFIRI. This combination is currently being evaluated in the Phase 2 DeFianCe study as a second-line treatment for patients with advanced microsatellite stable (MSS) colorectal cancer.
