Drug intelligence / Profile preview
A combination drug consisting of **sirolimus**, a small molecule mTOR inhibitor, and **panitumumab**, a recombinant fully human monoclonal antibody targeting the epidermal growth factor receptor (**EGFR**). Sirolimus inhibits the mammalian target of rapamycin pathway, leading to decreased cell proliferation and angiogenesis. Panitumumab binds to and blocks EGFR, preventing downstream cell signaling that promotes tumor growth. Used together, these agents aim to produce a synergistic antitumor effect by concurrently inhibiting the mTOR and EGFR pathways, both critical in cancer cell growth and survival. This combination has been investigated primarily in refractory solid tumors and metastatic colorectal cancer.
