Overview

**Sirpiglenastat + atezolizumab** is a combination investigational therapy consisting of DRP-104 (sirpiglenastat), a prodrug of the glutamine antagonist 6-diazo-5-oxo-L-norleucine (DON), and atezolizumab, an immune checkpoint inhibitor targeting the programmed death-ligand 1 (PD-L1). Sirpiglenastat antagonizes glutamine metabolism within tumors, leading to direct tumor cell cytotoxicity and immune modulation, including increased infiltration and activation of T cells, NK cells, and macrophages with decreased suppressive immune cells in the tumor microenvironment. Atezolizumab blocks PD-L1 to release inhibition on T lymphocyte-mediated tumor cell killing. This combination is under clinical investigation primarily for advanced solid tumors, including non-small cell lung cancer (NSCLC) and squamous cell carcinoma of the head and neck (SCCHN), to evaluate safety, anti-tumor activity, and synergistic immunomodulation[1][2][3][4][5][6].
