A combination therapy consisting of sirukumab, a human monoclonal antibody that selectively binds to and neutralizes interleukin-6 (IL-6), and methotrexate, a small molecule antimetabolite and immunosuppressant. Sirukumab is designed to block the pro-inflammatory effects of IL-6, a cytokine implicated in the pathogenesis of rheumatoid arthritis (RA). Methotrexate acts by inhibiting folate-dependent enzymes and modulating immune responses through multiple mechanisms, including adenosine accumulation and suppression of T and B cell function. This combination has been studied for the treatment of active rheumatoid arthritis, particularly in patients unresponsive to methotrexate or TNF inhibitors[1][3][4][5][7].
