Sitravatinib + tislelizumab is an investigational combination therapy for various advanced cancers. Sitravatinib is a spectrum-selective tyrosine kinase inhibitor targeting TAM receptors (TYRO3, AXL, MER), VEGFR-2, KIT, and MET. Tislelizumab is an anti-PD-1 monoclonal antibody. This combination aims to enhance antitumor immune responses by increasing tumor antigen presentation, promoting lymphocyte infiltration and migration, reducing myeloid-derived suppressor cells (MDSCs), and increasing the ratio of M1/M2-polarized macrophages. The combination has shown promising antitumor activity in multiple cancer types, including biliary tract cancer, non-small cell lung cancer, hepatocellular carcinoma, gastric/gastroesophageal junction cancer, and melanoma, particularly in patients with anti-PD-(L)1 resistant/refractory disease.
