Overview

SJ733 + tafenoquine is an investigational combination therapy being evaluated for the radical cure of uncomplicated Plasmodium vivax malaria. SJ733 is a small molecule dihydroisoquinolone that acts as a potent inhibitor of the Plasmodium P-type ATPase 4 (ATP4), a cation-transporting ATPase essential for maintaining sodium homeostasis within the parasite. Inhibition of ATP4 leads to a rapid influx of sodium, causing parasite swelling and clearance. Tafenoquine is an FDA-approved 8-aminoquinoline that provides radical cure by targeting the dormant liver stages (hypnozoites) of P. vivax, thereby preventing relapse. The combination is being developed to provide a simplified, short-course treatment regimen that addresses both blood-stage infection and liver-stage dormancy.
