Drug intelligence / Profile preview
SJP-0154 is a topical ophthalmic formulation of repaglinide, a small molecule ATP-sensitive potassium (K-ATP) channel blocker, currently in Phase 2 development by Senju Pharmaceutical for the treatment of dry eye disease. While repaglinide is traditionally utilized as an oral antidiabetic agent to stimulate insulin secretion by closing K-ATP channels in pancreatic beta cells, Senju is repurposing the compound for ocular use. The mechanism in dry eye involves the stimulation of tear secretion, potentially by modulating K-ATP channels in the lacrimal gland or ocular surface tissues to promote fluid release. This approach aims to increase tear volume and improve the clinical signs and symptoms of dry eye disease. SJP-0154 represents a novel application of a well-characterized systemic drug for a localized ophthalmic indication.
