SKB264 + inetetamab is an investigational combination therapy comprising the TROP2-directed antibody-drug conjugate (ADC) SKB264 (also known as sacituzumab tirumotecan or MK-2870) and the anti-HER2 monoclonal antibody inetetamab (Cipterbin). SKB264 consists of a humanized anti-TROP2 antibody conjugated to a belotecan-derivative topoisomerase I inhibitor payload (KL610023) via a pH-sensitive linker, allowing for targeted delivery and release within the tumor microenvironment. Inetetamab is a recombinant humanized monoclonal antibody targeting HER2, engineered with a modified Fc region to enhance antibody-dependent cell-mediated cytotoxicity (ADCC) compared to standard trastuzumab. This combination is being evaluated in the Phase II SHINE-HER trial for patients with HER2-positive, unresectable locally advanced or metastatic breast cancer who have progressed following treatment with trastuzumab deruxtecan (T-DXd).
