A combination therapy consisting of three agents: SKB264 (sacituzumab tirumotecan), an antibody-drug conjugate targeting TROP2; KL-A167 (tagitanlimab/Cotelet), a fully humanized monoclonal antibody targeting programmed cell death-ligand 1 (PD-L1); and cisplatin, a platinum-based chemotherapeutic. SKB264 is developed as a TROP2-targeting ADC delivering a cytotoxic payload to TROP2-positive tumor cells. KL-A167 blocks the PD-1/PD-L1 immune checkpoint to restore anti-tumor immunity. Cisplatin causes DNA crosslinking, triggering apoptosis in rapidly dividing cells. This combination is being evaluated primarily in advanced or metastatic non-small cell lung cancer and other solid tumors, aiming to synergize targeted killing, immune checkpoint inhibition, and cytotoxic chemotherapy[1][3][4][5][6].
