SL-701 + poly-ICLC is a combination immunotherapy regimen under investigation for the treatment of glioblastoma and other high-grade gliomas. SL-701 is a synthetic multi-peptide vaccine designed to induce a robust, target-specific anti-tumor immune response against key antigens overexpressed in glioblastoma, including interleukin-13 receptor alpha 2 (IL-13Rα2), ephrin A2 (EphA2), and survivin. These peptides are engineered to enhance immunogenicity and stimulate cytotoxic T-cell responses. Polyinosinic-polycytidylic acid stabilized with poly-L-lysine and carboxymethylcellulose (poly-ICLC) acts as an immune adjuvant, further boosting the immune system's response to the vaccine by mimicking viral double-stranded RNA and activating innate immunity via toll-like receptor 3 (TLR3). The combination has shown evidence of tumor regression and durable responses in phase 1/2 clinical trials for adults with recurrent glioblastoma as well as children with high-grade gliomas. Clinical studies have demonstrated that this therapy can generate strong CD8+ T-cell responses associated with improved survival outcomes[1][3][5][6][8].
