Overview

**SL-701 + poly-ICLC + bevacizumab** is an investigational combination therapy for recurrent glioblastoma multiforme (GBM). - **SL-701** is a synthetic multi-peptide cancer vaccine targeting tumor-associated antigens: survivin, interleukin-13 receptor alpha-2 (IL-13Rα2), and ephrin A2 (EphA2), all commonly overexpressed in glioma cells. It elicits a cytotoxic T-lymphocyte response against glioma cells presenting these antigens, promoting tumor cell destruction by the immune system[1][3][5][7]. - **Poly-ICLC** (polyinosinic-polycytidylic acid stabilized with poly-L-lysine and carboxymethylcellulose) is a synthetic double-stranded RNA that acts as a Toll-like receptor 3 (TLR3) agonist, functioning as an immunostimulant to boost the vaccine-induced immune response[5]. - **Bevacizumab** is a monoclonal antibody targeting vascular endothelial growth factor A (VEGFA), inhibiting angiogenesis and disrupting the tumor's blood supply, thereby starving tumor cells of oxygen and nutrients. It also potentially exerts direct cytotoxic effects on tumor cells by disrupting VEGFA-mediated survival and proliferation signaling[2][4][5][6]. This combination is designed to induce a targeted anti-tumor immune response while impairing the vascular support of the tumor.
