**SLNP@CpG** is a preclinical, locally administered lipid-nanoparticle formulation that encapsulates immunostimulatory CpG oligodeoxynucleotides. The CpG cargo activates Toll-like receptor 9 in tumor-associated macrophages, increasing phagocytosis and tumor-antigen cross-presentation, repolarizing immunosuppressive M2-like macrophages toward an M1-like state, and remodeling the tumor microenvironment toward antitumor immunity. In murine E.G7-OVA and B16-F10 melanoma models, intratumoral administration suppressed tumor growth, prolonged survival, and generated durable antitumor immune memory. The formulation was developed at Korea Advanced Institute of Science and Technology.
