Small interfering RNA targeting TBCE + cisplatin is an experimental combination therapy designed to overcome platinum-based resistance in hepatocellular carcinoma (HCC). The formulation consists of endosomal pH-responsive nanoparticles that co-deliver a small interfering RNA (siRNA) against tubulin folding cofactor E (TBCE) and the DNA-alkylating agent cisplatin. TBCE is a chaperone protein essential for tubulin heterodimer formation and microtubule dynamics; its overexpression is associated with poor prognosis and chemoresistance in liver cancer. By silencing TBCE, the therapy disrupts the cytoskeleton, thereby enhancing the sensitivity of tumor cells to cisplatin-induced DNA damage, cell cycle arrest, and apoptosis. This synergistic approach has demonstrated efficacy in reversing drug resistance in both orthotopic and patient-derived xenograft (PDX) models of liver cancer.
