Overview

**SMNP** is an investigational nanoparticle vaccine adjuvant composed of a saponin component, monophosphoryl lipid A, cholesterol, and phospholipid; clinical-grade material has been reported to use QS-21 saponin and dipalmitoylphosphatidylcholine. MPLA provides Toll-like receptor 4 stimulation, while the saponin-containing nanoparticle promotes antigen transport to draining lymph nodes and supports innate activation, germinal-center responses, T-cell help, and durable antibody and memory-B-cell responses. SMNP contains no HIV antigen itself; it is being evaluated as the adjuvant administered with the germline-targeting HIV envelope trimer N332-GT5 gp140 in the Phase 1 HVTN 144 study in adults without HIV.
