SMT-NK + pembrolizumab is a combination investigational cancer immunotherapy consisting of highly activated allogeneic natural killer (NK) cells (SMT-NK) and the anti-PD-1 monoclonal antibody pembrolizumab. SMT-NK, developed by SMT Bio, is an off-the-shelf NK cell therapy designed to enhance innate immune responses against tumor cells. Pembrolizumab (Keytruda), developed by Merck & Co., is a checkpoint inhibitor that blocks the programmed death receptor 1 (PD-1) on T-cells, preventing tumor-induced immune suppression and enabling T-cell mediated antitumor activity. The combination aims to synergistically boost both innate and adaptive immunity for improved efficacy in treating advanced biliary tract cancer, particularly in patients refractory to standard chemotherapy. Clinical trials have shown that this regimen can be safely administered with no severe adverse events directly related to the combination and has demonstrated improved antitumor activity compared to monotherapy with pembrolizumab[3][5][6][8].
