SNK01 + avelumab + pembrolizumab is an investigational combination therapy regimen developed by NKGen Biotech. It pairs SNK01—an autologous natural killer (NK) cell therapy consisting of patient-derived NK cells expanded and activated for high cytotoxicity—with the immune checkpoint inhibitors avelumab (targeting PD-L1) or pembrolizumab (targeting PD-1). The rationale for the combination is to utilize the direct tumor-killing capabilities of activated NK cells while simultaneously blocking the PD-1/PD-L1 pathway, which tumors often use to evade immune detection. This approach is intended to treat patients with advanced or metastatic refractory solid tumors, including those that are PD-L1 negative. Clinical development is currently in Phase 1, with specific studies focusing on non-small cell lung cancer (NSCLC) and sarcomas.
