SNK01 is an autologous natural killer (NK) cell therapy developed by NKMax (NKGen Biotech). It consists of non-genetically modified autologous NK cells that have been expanded and activated to exhibit enhanced cytotoxicity. This specific regimen combines SNK01 with the chemotherapy agents gemcitabine and cisplatin, along with the anti-EGFR monoclonal antibody cetuximab (Erbitux). It is primarily investigated for the treatment of patients with non-small cell lung cancer (NSCLC) who have failed prior tyrosine kinase inhibitor (TKI) therapy, particularly those with EGFR mutations. The mechanism involves the direct anti-tumor activity of the NK cells, which is further augmented by cetuximab-mediated antibody-dependent cellular cytotoxicity (ADCC), alongside the cytotoxic effects of the chemotherapy doublet. Clinical data from Phase I/IIa trials (NCT04872634) demonstrated that the combination was safe and showed potential antitumor activity, with an objective response rate of 25% and a disease control rate of 100%.
