SNS-101 + cemiplimab is a combination therapy under clinical investigation for the treatment of advanced solid tumors, particularly in patients who are resistant to or have progressed on prior anti-PD-1/PD-L1 immunotherapies. SNS-101 (also known as solnerstotug) is a pH-selective, human monoclonal antibody that targets V-domain Ig suppressor of T-cell activation (VISTA), a negative checkpoint regulator primarily expressed on myeloid cells. By neutralizing VISTA, SNS-101 aims to alleviate this immunosuppressive pathway and promote anti-tumor immune responses. Cemiplimab is a fully human, IgG4 monoclonal antibody that blocks the programmed cell death protein 1 (PD-1) checkpoint on T cells; it is an FDA-approved immune checkpoint inhibitor marketed as Libtayo. The combination aims to synergistically enhance immune activation against tumor cells. Early clinical data indicate a tolerable safety profile for both monotherapy and combination arms, with preliminary evidence of clinical activity in resistant tumors[1][2][3][5].
