SNX-5422 + everolimus

SNX-5422 + everolimus is an investigational oral combination regimen studied for the treatment of advanced neuroendocrine tumors (NETs). SNX-5422 is a pro-drug of SNX-2112, a highly potent, selective small-molecule inhibitor of heat shock protein 90 (Hsp90), which plays a key role in stabilizing multiple proteins involved in cancer cell survival and proliferation. Everolimus is an allosteric inhibitor of the mammalian target of rapamycin (mTOR), a key regulatory serine/threonine kinase that controls cell growth, proliferation, and survival. Preclinical studies suggested additive antitumor effects from the combined inhibition of Hsp90 and mTOR. The combination was evaluated in a phase 1, open-label, dose-escalation clinical trial in subjects with unresectable neuroendocrine tumors[1][2][3][5][7].
