Overview

sodium (3-oxo-(17r)-17-28-norlup-20(29)-en)-2-oxoethyl-phosphonate is a semisynthetic pentacyclic triterpenoid derivative based on the betulonic acid (BoA) skeleton. It features a methylphosphonate moiety at the C17 position, which was designed to improve the pharmacokinetic and pharmacodynamic profile of natural triterpenoids like betulinic acid. In preclinical studies, the compound has demonstrated significant antimelanoma activity, specifically showing dose-dependent antiproliferative and anti-migratory effects against the B164A5 murine melanoma cell line. Mechanistically, it appears to induce apoptosis, as evidenced by nuclear condensation, while maintaining a favorable safety profile with low cytotoxicity to normal cells and no evidence of vascular disintegration in HET-CAM assays.
