Overview

Sofosbuvir + daclatasvir is a fixed-dose oral combination of two direct-acting antiviral agents used for the treatment of chronic hepatitis C virus (HCV) infection. Sofosbuvir is a nucleotide analog inhibitor targeting the HCV NS5B RNA-dependent RNA polymerase, thereby inhibiting viral replication. Daclatasvir is an inhibitor of the HCV NS5A protein, which plays a key role in viral replication and assembly. This combination offers high efficacy across all major HCV genotypes (1–6), with sustained virologic response rates exceeding 90% in most patient populations, including those with cirrhosis or HIV coinfection. The regimen is typically administered once daily for 12 weeks and may be extended or combined with ribavirin in certain cases such as compensated cirrhosis or prior treatment failure. It has also been investigated for use against COVID-19 due to similarities between hepatitis C and SARS-CoV-2 viruses[1][4][6].
