Overview

This is an investigational oral antiviral combination therapy consisting of three direct-acting antivirals targeting hepatitis C virus (HCV): sofosbuvir, daclatasvir, and CDI-31244. Sofosbuvir is a nucleotide analog inhibitor of the HCV NS5B RNA-dependent RNA polymerase. Daclatasvir is an inhibitor of the HCV NS5A protein. CDI-31244 (also called CC-31244) is a non-nucleoside inhibitor (NNI) that targets the HCV NS5B polymerase at a different site from sofosbuvir and has broad-spectrum activity against all major HCV genotypes with a high barrier to resistance[8][10]. The rationale behind this triple regimen is to achieve ultrashort duration therapy while maintaining high cure rates by combining agents with complementary mechanisms and resistance profiles.
