**sofosbuvir + GS-0938** is a combination of two nucleotide analogue inhibitors of the hepatitis C virus (HCV) NS5B RNA-dependent RNA polymerase. **Sofosbuvir** is a pyrimidine nucleotide analogue, and **GS-0938** is a purine nucleotide analogue. Both act as direct-acting antivirals (DAAs) and work by blocking viral replication. The combination was developed for oral, interferon-free therapy in chronic HCV infection and has shown substantial reductions in viral load and high effectiveness for reducing HCV RNA levels in phase 2 trials in treatment-naïve HCV genotype 1 patients; both agents demonstrated additive antiviral effects. No viral breakthroughs or resistance were observed in short-term studies[1][3][2]. Developed primarily by Gilead Sciences for HCV infection, this investigational combination was well tolerated in clinical trials, with no serious adverse events reported[1][2][3]. As both are nucleotide analogues targeting the same viral polymerase, further development focused on combining sofosbuvir with other classes of antivirals for greater efficacy.
