Overview

Sofosbuvir + ribavirin is a fixed combination antiviral regimen used for the treatment of chronic hepatitis C virus (HCV) infection. Sofosbuvir is a nucleotide analog NS5B polymerase inhibitor that blocks HCV RNA replication, while ribavirin is a nucleoside analog with broad-spectrum antiviral activity, thought to enhance viral clearance through multiple mechanisms including inhibition of viral RNA synthesis and immunomodulation. This combination provides an interferon-free option for patients with certain HCV genotypes, especially those who are ineligible or intolerant to interferon-based therapies. The regimen has demonstrated efficacy in both treatment-naive and experienced patients, including those with compensated cirrhosis or post-liver transplantation[1][2][4][6]. It is generally well tolerated; common adverse effects include fatigue, headache, anemia, diarrhea, and nausea[1][4].
