This is a combination antiviral regimen consisting of three direct-acting antivirals used for the treatment of chronic hepatitis C virus (HCV) infection, particularly genotype 1. Sofosbuvir is a nucleotide analog NS5B polymerase inhibitor that blocks HCV RNA replication. Simeprevir is an NS3/4A protease inhibitor that prevents viral polyprotein processing and replication. Ribavirin is a nucleoside analog with broad-spectrum antiviral activity, often used to enhance the efficacy of other antivirals in HCV therapy. This triple combination has been studied in both treatment-naive and experienced patients, including those who have failed previous therapies. The regimen may be considered for patients unable to tolerate interferon or as part of all-oral therapy options[1][5][6]. The addition of ribavirin can increase sustained virologic response rates in certain populations.
