Drug intelligence / Profile preview
This is a **combination therapy consisting of a soluble beta-glucan (a natural immunomodulatory polysaccharide), a standard antibody (typically a tumor-directed monoclonal antibody), and chemotherapy**. Soluble beta-glucan acts as an immune adjuvant, priming and activating innate immune cells (such as macrophages and neutrophils) through receptors like Dectin-1 and CR3, and can convert these cells into antitumor effectors. In this combination, the antibody opsonizes tumor cells and activates the complement system, leading to iC3b coating of tumor cells. Beta-glucan primes leukocytes to recognize these coated tumor cells, enhancing targeted cytotoxicity through production of proinflammatory cytokines and involving both innate and adaptive immune arms. Chemotherapy works independently via direct cytotoxic mechanisms and also augments immunogenic cell death, providing further synergy with the immune-stimulatory effects of the other agents. This approach is under investigation for various cancers, including gastric adenocarcinoma and colorectal cancer, showing enhanced objective response rates and manageable safety profiles, particularly in the context of first-line therapy. Multiple beta-glucan formulations are under clinical development, often combined with monoclonal antibodies such as camrelizumab, cetuximab, or immune checkpoint inhibitors[1][2][3][5].
