This combination therapy regimen involves the concomitant administration of somatropin (recombinant human growth hormone), leuprolide (a GnRH agonist), and levothyroxine (thyroid hormone) to improve the final adult height of adolescent patients with long-term acquired hypothyroidism. In these patients, standard thyroid hormone replacement therapy can sometimes lead to rapid bone maturation and premature epiphyseal closure, which limits growth potential. Somatropin is utilized to restore and stimulate growth velocity, while leuprolide is employed to delay puberty and prevent the early closure of growing bones, thereby extending the growth window. Levothyroxine serves as the primary replacement therapy for the underlying thyroid deficiency. This specific therapeutic approach was evaluated in a Phase 4 clinical trial led by the Baylor College of Medicine.
