Overview

Sonepcizumab + ranibizumab is a combination of two biologic drugs under investigation primarily for the treatment of neovascular eye diseases, including neovascular age-related macular degeneration (nAMD) and polypoidal choroidal vasculopathy (PCV)[1][5]. Sonepcizumab is a humanized monoclonal antibody targeting sphingosine-1-phosphate, which is implicated in angiogenesis and vascular leakage, although clinical development substantially focused on oncology and ophthalmology. Ranibizumab is a recombinant humanized monoclonal antibody fragment (Fab) that inhibits vascular endothelial growth factor A (VEGF-A), a key driver of pathological blood vessel growth and increased vascular permeability in ocular diseases[4][6]. The rationale for combination therapy includes broader inhibition of angiogenic pathways and a potential for superior anatomic and functional outcomes compared to anti-VEGF-A monotherapy[1][5].
