**Sonidegib + buparlisib** is an investigational oral targeted-therapy combination developed by Novartis. Sonidegib is a small-molecule Smoothened antagonist that suppresses oncogenic Hedgehog signaling, while buparlisib is a pan-class I phosphatidylinositol 3-kinase inhibitor that suppresses PI3K/AKT pathway signaling. The agents were co-administered as separate oral medicines in a Phase 1b dose-escalation and expansion study in advanced solid tumors, including metastatic breast cancer, pancreatic adenocarcinoma, metastatic colorectal cancer, and recurrent glioblastoma, and in a Phase 2 pilot study in advanced or metastatic basal cell carcinoma.
