Sonidegib (LDE225) is a selective oral small molecule inhibitor of Smoothened, a key component of the Hedgehog signaling pathway. Etoposide is a topoisomerase 2A inhibitor that interferes with DNA replication and cell division. Cisplatin is a platinum-based chemotherapeutic agent that causes DNA crosslinking and apoptosis in cancer cells. The combination of sonidegib with etoposide and cisplatin has been investigated in clinical trials for extensive-stage small cell lung cancer (ES-SCLC). Sonidegib targets tumor growth driven by aberrant Hedgehog pathway activation; etoposide and cisplatin are standard first-line cytotoxic agents for SCLC[1][2][6].
