Overview

Sonidegib (LDE225) is a small molecule inhibitor of the Hedgehog signaling pathway that acts by selectively inhibiting Smoothened (SMO), a key transmembrane protein in the pathway. It is developed for targeting tumor stroma and has been studied in combination with the chemotherapy agent gemcitabine for advanced and metastatic pancreatic ductal adenocarcinoma (PDAC). Gemcitabine is a nucleoside analog used as chemotherapy. The combination aims to disrupt tumor-stromal interactions and inhibit cancer cell proliferation by blocking aberrant Hedgehog signaling while simultaneously exerting cytotoxic effects through DNA synthesis inhibition[1][2][5][6]. Sonidegib was developed by Novartis.
