Overview
Sonidegib (LDE225) is an orally bioavailable small molecule inhibitor of the Smoothened (SMO) receptor, a key component of the Hedgehog (Hh) signaling pathway. Paclitaxel is a taxane-based chemotherapy agent that stabilizes microtubules, thereby interfering with cell division and inducing apoptosis. The combination of sonidegib and paclitaxel has been investigated in Phase I clinical trials, notably by the Swiss Group for Clinical Cancer Research (SAKK), for the treatment of advanced solid tumors and platinum-resistant ovarian cancer. The rationale for this combination is to target the Hedgehog pathway, which is often aberrantly activated in various cancers and contributes to tumor progression, maintenance of cancer stem cells, and resistance to conventional chemotherapy.
