Overview

Sorafenib + dacarbazine is a combination therapy used primarily in the treatment of advanced or metastatic melanoma. Sorafenib is an oral small molecule multi-kinase inhibitor that targets several kinases involved in tumor cell proliferation and angiogenesis, including RAF kinases (such as BRAF), vascular endothelial growth factor receptors (VEGFR), and platelet-derived growth factor receptor (PDGFR). Dacarbazine is an alkylating agent that works by methylating DNA and thereby inhibiting DNA synthesis and function. The combination has been studied in multiple phase I/II clinical trials for metastatic melanoma, showing acceptable toxicity profiles and some antineoplastic activity. Sorafenib may also have immunomodulatory effects when combined with dacarbazine[1][2][3][4][5][6].
