Overview

Sorafenib + interferon is a combination therapy consisting of the small molecule multikinase inhibitor sorafenib and an immunomodulatory cytokine, typically interferon alpha (either alpha-2a or alpha-2b). Sorafenib inhibits tumor cell proliferation by blocking the RAF/MEK/ERK pathway and reduces angiogenesis by inhibiting vascular endothelial growth factor receptor (VEGFR) and platelet-derived growth factor receptor (PDGFR) signaling. Interferon-alpha exerts antitumor effects primarily through antiangiogenic activity and immune regulation. This combination has been investigated in clinical trials for advanced renal cell carcinoma (RCC) and melanoma, showing preliminary antitumor activity with manageable toxicity profiles[1][2][5]. The combination is not approved as a standard regimen but has demonstrated efficacy in phase I/II studies.
