Overview

Sorafenib + nivolumab is a **combination of two pharmaceutical drugs**: sorafenib, a small molecule multikinase inhibitor, and nivolumab, a monoclonal antibody that is a programmed death-1 (PD-1) immune checkpoint inhibitor. Sorafenib is primarily used as a first-line systemic therapy for advanced hepatocellular carcinoma (HCC)[2], while nivolumab is indicated for multiple cancers as an immunotherapy. Combined use is being investigated for potential **synergistic effects in advanced HCC**, especially in patients refractory or intolerant to sorafenib, as sorafenib targets tumor cell proliferation and angiogenesis, and nivolumab stimulates antitumor immune response by inhibiting PD-1-mediated T-cell suppression. Clinical evidence, including case reports and retrospective studies, suggests that this combination may enhance efficacy compared to single-agent therapies, but it is not currently an FDA-approved combination regimen for any indication.
