Overview

Sorafenib + vinorelbine is an investigational combination therapy consisting of two small molecule drugs, sorafenib and vinorelbine. Sorafenib is a multikinase inhibitor that targets several kinases involved in tumor cell proliferation and angiogenesis, including RAF kinases, vascular endothelial growth factor receptors (VEGFR), and platelet-derived growth factor receptor (PDGFR). Vinorelbine is a semi-synthetic vinca alkaloid that inhibits microtubule assembly by binding to tubulin, thereby disrupting mitosis and inducing apoptosis in cancer cells. The combination has been studied for its potential synergistic antitumor effects in preclinical models of hepatocellular carcinoma (HCC) as well as in phase I/II clinical trials for metastatic breast cancer. Preclinical studies have shown enhanced efficacy compared to monotherapy, with increased tumor necrosis and apoptosis. Clinical trials indicate the combination is feasible but associated with notable toxicity due to pharmacokinetic interactions[1][2][3].
