Overview

Sorafenib + vitamin K1 is an experimental combination therapy investigated for its synergistic effects in treating various cancers, including hepatocellular carcinoma (HCC), pancreatic adenocarcinoma, and malignant glioma. Sorafenib is a multi-kinase inhibitor that targets the RAF/MEK/ERK signaling pathway, while vitamin K1 (phylloquinone) has been shown to enhance sorafenib's growth-inhibitory and pro-apoptotic effects. Preclinical studies indicate that this combination can achieve therapeutic efficacy at lower doses of sorafenib, potentially reducing the drug's systemic toxicity. The mechanism involves the simultaneous inhibition of the RAF/MEK/ERK pathway and the activation of the JNK/c-Jun/FasL apoptotic pathway.
