sorbitol + tislelizumab + oxaliplatin + S-1 is an experimental combination regimen being evaluated for the neoadjuvant treatment of locally advanced gastric cancer and gastroesophageal junction adenocarcinoma. The regimen combines the PD-1 inhibitor tislelizumab with the SOX chemotherapy backbone (comprising the oral fluoropyrimidine S-1 and intravenous oxaliplatin) and oral sorbitol. Sorbitol is included as a metabolic modulator intended to enhance the efficacy of the chemo-immunotherapy combination. Tislelizumab is a humanized monoclonal antibody that blocks the interaction between PD-1 and its ligands, thereby restoring T-cell mediated anti-tumor activity. The SOX regimen is a standard-of-care chemotherapy in East Asian populations, providing cytotoxic effects through DNA cross-linking and inhibition of thymidylate synthase.
