Overview

Sotorasib + atezolizumab is a combination therapy comprising **sotorasib**, a targeted small molecule inhibitor of the mutant KRAS G12C protein, and **atezolizumab**, a monoclonal antibody checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1). Sotorasib binds covalently to the KRAS G12C mutant protein, locking it in an inactive state and thereby inhibiting oncogenic signaling. Atezolizumab blocks the interaction of PD-L1 with its receptors PD-1 and B7.1, enhancing T-cell mediated anti-tumor immune responses. This combination has been investigated in clinical trials for the treatment of advanced non-small cell lung cancer (NSCLC) with KRAS G12C mutations, aiming to improve antitumor activity through complementary mechanisms of direct oncogenic inhibition and immune checkpoint blockade[1][2].
