Overview

Sotorasib + bevacizumab is a combination therapy involving sotorasib, a first-in-class small molecule that irreversibly inhibits the KRAS G12C mutant protein, and bevacizumab, a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A). Sotorasib targets the KRAS G12C mutation found in certain cancers, including metastatic colorectal cancer and non-small cell lung cancer, blocking oncogenic signaling and leading to tumor suppression. Bevacizumab inhibits VEGF-mediated angiogenesis, thereby reducing tumor blood supply and growth. The combination aims to target tumor growth from two mechanistic pathways: direct inhibition of a key oncogenic driver and shutdown of angiogenesis. Developers of sotorasib include Amgen, while bevacizumab was developed by Genentech. Combination use of sotorasib with bevacizumab is being evaluated in clinical studies for first-line therapy of KRAS G12C-mutated metastatic colorectal cancer, offering a targeted and biomarker-driven approach designed to improve patient outcomes compared to current standards of care[3].
