Sotorasib + bevacizumab is a combination therapy involving sotorasib, a first-in-class small molecule that irreversibly inhibits the KRAS G12C mutant protein, and bevacizumab, a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A). Sotorasib targets the KRAS G12C mutation found in certain cancers, including metastatic colorectal cancer and non-small cell lung cancer, blocking oncogenic signaling and leading to tumor suppression. Bevacizumab inhibits VEGF-mediated angiogenesis, thereby reducing tumor blood supply and growth. The combination aims to target tumor growth from two mechanistic pathways: direct inhibition of a key oncogenic driver and shutdown of angiogenesis. Developers of sotorasib include Amgen, while bevacizumab was developed by Genentech.
