Overview

sotorasib + chemotherapy is a combination therapy regimen developed by Amgen for the treatment of advanced solid tumors characterized by the KRAS G12C mutation. The chemotherapy components evaluated in clinical trials like CodeBreaK 101 include platinum-based agents (carboplatin, oxaliplatin), antimetabolites (5-fluorouracil, pemetrexed), taxanes (docetaxel, paclitaxel), and topoisomerase inhibitors (irinotecan). By combining targeted KRAS G12C inhibition with the broad cytotoxic effects of chemotherapy, the regimen aims to enhance anti-tumor efficacy, achieve deeper responses, and delay the emergence of resistance in patients with KRAS G12C-mutated cancers such as non-small cell lung cancer and colorectal cancer.
