This is a combination regimen consisting of **sotorasib**, **panitumumab**, and the **FOLFIRI** chemotherapy protocol. - **Sotorasib** is a small molecule inhibitor that targets the KRAS G12C mutation, leading to selective, irreversible inhibition of mutant KRAS protein activity, which results in suppression of oncogenic signaling and inhibition of tumor cell growth. - **Panitumumab** is a fully human monoclonal antibody targeting the epidermal growth factor receptor (EGFR), inhibiting EGFR signaling downstream, which disrupts cell proliferation and survival in EGFR-expressing cancer cells. - **FOLFIRI** is a three-drug chemotherapy regimen composed of folinic acid (leucovorin), fluorouracil (5-FU), and irinotecan. Folinic acid enhances the action of fluorouracil, which is a thymidylate synthase inhibitor and antimetabolite interfering with DNA synthesis; irinotecan is a topoisomerase I inhibitor, blocking DNA replication and leading to cell death. This combination is investigated or considered for use in **RAS wild-type** or **KRAS-mutated** advanced colorectal cancer (most notably metastatic colorectal cancer) as a targeted plus cytotoxic therapy strategy.
