Drug intelligence / Profile preview
Lumakras (sotorasib) in combination with platinum doublet chemotherapy is an investigational combination regimen being evaluated for the treatment of KRAS G12C-mutated advanced or metastatic non-small cell lung cancer (NSCLC). Sotorasib is a small molecule that specifically and irreversibly inhibits the KRAS G12C mutant protein by locking it in an inactive GDP-bound state. Platinum doublet chemotherapy typically consists of a platinum agent (such as carboplatin or cisplatin) combined with a doublet partner (such as pemetrexed), which works by damaging DNA and inhibiting cell division. This combination regimen is designed to enhance anti-tumor efficacy and delay or overcome the emergence of resistance pathways in KRAS G12C-mutated NSCLC. It is currently being evaluated in clinical trials, including the Phase 3 CodeBreaK 202 study as a first-line therapy for patients with PD-L1-negative, advanced KRAS G12C-mutated NSCLC.
