sotorasib + trametinib + panitumumab

This drug is a combination therapy consisting of **sotorasib**, **trametinib**, and **panitumumab**, used in investigational treatment regimens for advanced solid tumors, primarily metastatic colorectal cancer (CRC) with a **KRAS G12C mutation**. Sotorasib is a small molecule inhibitor targeting KRAS G12C, a prevalent mutation in CRC and non-small cell lung cancer. Trametinib is a MEK inhibitor that blocks downstream signaling from mutant RAS proteins. Panitumumab is a fully human monoclonal antibody targeting and inhibiting the epidermal growth factor receptor (EGFR). The rationale for combining these agents is to inhibit multiple pathways involved in tumor growth and resistance, improving response and delaying or overcoming resistance seen with single-agent targeted therapies. Clinical trials, especially the CodeBreaK 101 sub-cohort, have investigated the use of these drugs together in patients with various advanced KRAS G12C–mutated solid tumors, including metastatic colorectal cancer and non-small cell lung cancer.
