**Spartalizumab + everolimus** is an experimental drug combination consisting of spartalizumab, a humanized monoclonal antibody targeting the programmed death-1 (PD-1) receptor, and everolimus, an orally available mTOR inhibitor. Spartalizumab acts as an immune checkpoint inhibitor by blocking PD-1 on T cells, enhancing anti-tumor immune responses. Everolimus inhibits the mammalian target of rapamycin (mTOR), reducing cell proliferation and angiogenesis. The combination aims to synergistically promote anti-cancer immune activity while directly inhibiting tumor cell survival and growth. This strategy is under early-phase (phase I/II) clinical investigation for advanced solid tumors including melanoma and other cancers, with the goal of improving outcomes through dual immunomodulatory and targeted mechanisms.
