Overview

A combination regimen consisting of **spartalizumab** (a humanized monoclonal antibody targeting Programmed Death-1 [PD-1]), **paclitaxel** (a microtubule-stabilizing cytotoxic chemotherapeutic agent), and **carboplatin** (a platinum-based DNA-damaging cytotoxic chemotherapeutic). This combination is under clinical investigation for use in advanced solid tumors such as non-small cell lung cancer (NSCLC) and potentially other cancers. Spartalizumab blocks the interaction between PD-1 and its ligands (PD-L1/PD-L2), thereby enhancing antitumor T cell responses by relieving PD-1 mediated immune suppression. Paclitaxel inhibits cell division by stabilizing microtubules and preventing their depolymerization, while carboplatin promotes DNA crosslinking and damages tumor cell DNA, resulting in cell death. Preclinical and early phase clinical results suggest this combination is tolerable and shows activity in platinum-doublet chemotherapy regimens for cancers including NSCLC[1][8].
