Overview
This is an investigational combination regimen composed of four agents: spartalizumab (a monoclonal antibody against PD-1), pemetrexed (an anti-folate antineoplastic chemotherapy), carboplatin (a platinum-based chemotherapy), and canakinumab (a monoclonal antibody targeting interleukin-1β). Spartalizumab and canakinumab act as immune-modulating agents; pemetrexed and carboplatin act as cytotoxic agents targeting rapidly dividing cells. This combination is explored in advanced non-small cell lung cancer (NSCLC), aiming to leverage dual immune checkpoint blockade alongside chemotherapy for enhanced antitumor efficacy. Spartalizumab is being developed by Novartis for immunotherapy indications; pemetrexed is widely approved for NSCLC and developed by Eli Lilly, carboplatin was developed by Bristol Myers Squibb, and canakinumab is developed by Novartis and approved for autoinflammatory diseases, with cancer applications under investigation.
