Overview

This is a **combination therapy** comprising four active pharmaceutical ingredients: **spartalizumab** (a monoclonal antibody against PD-1), **pemetrexed** (a folate antimetabolite chemotherapy), **cisplatin** (a platinum-based chemotherapy), and **canakinumab** (an IL-1β targeting monoclonal antibody). Spartalizumab inhibits the PD-1/PD-L1 immune checkpoint, enhancing T-cell mediated antitumor immune responses. Pemetrexed and cisplatin are cytotoxic agents that inhibit DNA synthesis and induce DNA damage, leading to tumor cell death. Canakinumab inhibits IL-1β signaling, modifying inflammatory responses; its rationale in cancer therapy includes reducing tumor-promoting inflammation. This combination is being investigated in metastatic non-small cell lung cancer (NSCLC), particularly in populations unselected for PD-L1 expression.
