spesolimab + placebo

Spesolimab is a high-affinity, specific, humanized IgG1 monoclonal antibody that antagonizes the interleukin-36 receptor (IL-36R) by binding at a site distinct from its agonists IL-36α/β/γ and endogenous antagonists. It inhibits IL-36R-mediated signaling pathways involved in inflammatory diseases such as generalized pustular psoriasis (GPP), palmoplantar pustulosis (PPP), hidradenitis suppurativa, and Netherton syndrome. Spesolimab reduces T-cell stimulation and pro-inflammatory processes by blocking the IL-36 axis. It was developed primarily for GPP flares treatment and has demonstrated rapid and sustained clinical improvements with a favorable safety profile. The drug was approved by the FDA in September 2022 for GPP flares in adults.[2][4][5][6]
